Summary: Recent large-scale studies increasingly illustrate the phenotypic changes monitored by RNA-Seq, which--in contrast to the genotype--also rely on the repertoire of genes expressed in a respective cell type or state. Our journey takes us to different types of transcriptomic variation observed between human tissues and individuals, from the same and from different populations respectively ethnic groups, their genetic origins and some of their functional impacts as by splicing and poly-adenylation. We have a look at the changes of the mutual interplay of the nuclear and the mitochondrial transcriptome in some of these individual-specific cell states, and sketch chances and challenges provided by 3rd generation sequencing technologies to assess new aspects of transcriptome variation.
